报告摘要

The therapeutic action of traditional low molecular weight drugs is often hampered due to side effects that are the result of off-target delivery, relatively low dosages, which are related to limited solubilities in physiologically relevant media, as well as difficulties to deliver these actives across physiological barriers such as the blood brain barrier.

The use of polymer-based therapeutics and the use of nano-sized polymer delivery systems allow to overcome some of the drawbacks. The interest in using polymers as therapeutics or as carrier systems goes back to the early 1970’s. In particular over the past decade or so there has been an explosive growth in research activities in this area. A lot of recent research is driven on the one hand by chemistry, e.g. the expanding arsenal of chemoselective (and responsive and reversible) conjugation chemistries that are nowadays available, but also due to spectacular advances in instrumentation, in particular microscopy that now allow to visualize biological processes in live cells on unprecedented length scales. Furthermore, great advances have been made in our understanding of interactions between cells and synthetic materials as well as in the ability of researchers to develop more reliable in vitro and in vivo models to evaluate polymer therapeutics.

This lecture will start with a brief introduction into the history and origins of the field of polymer nanomedicine and discuss some of the lessons learned from first clinical trials that were carried out with polymer – drug conjugates in the 1990’s. After that, some of the current challenges and opportunities for this field will be presented. Using examples from our own research, some showcases will be presented that illustrate how the use of polymers and polymer nanocarriers allows to address some of these challenges. This includes the use of polymers and polymer-peptide conjugates to combat viral infections, the development of multifunctional polymer conjugates that allow spatiotemporal control release to overcome drug resistance as well as the use of isotopically or trace-element labelled polymers to investigate cell uptake and internalization without the need for fluorescent labels. As a final example, the potential of pro- and eukaryotic cells as carriers for the delivery of polymers and polymer nanomedicines will be presented.

 

报告人简介:

Prof. Harm-Anton Klok studied chemical technology at the University of Twente (The Netherlands) from 1989 to 1993 and received his Ph.D. in 1997 from the University of Ulm. After postdoctoral research with David N. Reinhoudt (University of Twente) and Samuel I. Stupp (University of Illinois at Urbana–Champaign, USA), he joined the Max Planck Institute for Polymer Research (Germany) in early 1999 as a project leader in the group of Klaus Müllen. In November 2002, he was appointed to the faculty of EPFL. Since 2012 he is Director of the Institute of Materials (~ Department Head) and also directs the Molecular and Hybrid Materials Characterization Center at EPFL. His research interests include polymer nanomedicine as well as polymer surface and interface science.

Prof. Harm-Anton Klok is recipient of the Arthur K. Doolittle Award of the American Chemical Society (2007) and is Associate Editor of the American Chemical Society journal Biomacromolecules and serves or has served on the editorial advisory board of Eur. Polym. J., J. Polym. Sci. A: Polym. Chem., Macromolecules, ACS Macro Letters as well as Macromol. Rapid Commun., Macromol. Bioscience and Chimia. He has been a Visiting Professor at the University of Bordeaux (France) and the University of Massachusetts/Amherst (USA), is a Chair Professor at Soochow University (Suzhou, China), guest professor at the Changchun Institute of Applied Chemistry, Chinese Academy of Sciences, Shanghai University and Nanyang Technological University (Singapore) and was awarded a Chinese Academy of Sciences visiting professorship for senior international scientists (Institute of Chemistry, Chinese Academy of Sciences, Beijing).
